My ex & I will be going to court for custody issues. I recently found out his "attorney" is his aunt, a pre-law graduate specializing 100% in Criminal Defense. I know no one can KNOW how much she knows about family law, but in general do criminal defense attorneys really know enough? I've heard for custody matters it's fairly imperative to have an attorney specializing in Family Law. Just wanting to know what I'm up against.

there is no pre-law major (she most likely majored in history, politics, english, or liberal arts)...therefore, at this stage, she doesn't know jack about much at all in the legal arena...besides which, she can't legally represent him in court without a license to practice law in your state.

It's not really clear where she is in her studies, but she's not an attorney and can't represent him in court. If she's really studying law (like at a law school), she'd be exposed to some family law, and related issues and procedures are also going to come up in her criminal studies. She also may have worked some place where she's been around such things.
But if she's doing something like a "criminal justice" major at one of those for-profit schools, she's not really going to know her way around family issues to any significant degree.
Anyway, you might want to make sure what she's studying. It doesn't sound like she's even studying to be a criminal defense "attorney."
