Last night Dow was about worthless, and today they're about 27,000 points up. Does that mean that, had I bought $1000 of stock last night, that it would be worth $27,000 today? How accessible are is stock investments?

Dow is an index of the top 30 industrial stocks - and it was not worthless 'last night'. Dow is not a stock.

Does not mean that at all. The DOW is a composite index and does not reflect the value of one individual sock. Your imaginary stock could even have dropped significantly even if the overall index went up.
Where in the world were you looking that said it was about worthless? It closed yesterday at 27,691.50. And no, the gain or loss on any individual stock does NOT equal the amount of change in the DOW. The DOW can go up, your stock can go down, or vice versa.
