There is no reason for the comma after "therefore."

The phrase you're using, in American English anyway, is "calls the shots," meaning to be in charge, one's judgment unquestioned. (Sounds just like God to me.) If a person calls *his* shots, he is predicting the outcome of those shots, often in a game of pool. ("Nine ball in the corner pocket, six on the side" is calling his shots.)

So you probably want to write "he calls the shots."

It's customary to capitalize pronouns referring to God, when you mean the Christian God, so you'd write: "...therefore He calls the shots."
