I hate bust in on a question, but I haven't seen a good answer yet. The drive isn't as bad as most would make it.
My wife & I drove from Brooksville, FL to Alaska and back, a trip exceeding 16,000 mile, in a motorhome. The roads are fine, if you see a sign that indicates a bump, you had better slow down to as slow a speed as you can. The have frost heaves that will buckle the road as high as a foot in spots.
Once in Alaska you can drive to all the major cities except the capital, Juneau. By boat or plane is the only way to the capital. Fairbanks, Anchorage, Wasilla, Skagway, Haines, Seward to name a few.
Before taking the trip you should purchase a copy of the Milepost, the Alaskan travelers bible. anything along the Alaskan Highway is noted. Even side trips from Whitehorse, Yukon Territory to Skagway, Alaska. This book can be purchased from the Milepost or Amazon.
Visit my Yahoo 360 Blog for photos along the road from Dawson Creek, British Columbia, the start of the Alaskan Highway to Delta Junction, the end of the Alaskan Highway. The road goes on to Fairbanks and beyond.
Enjoy
While I was composing my answer someone actually answered who had actually been on the road, sorry

As far as going to the more populated parts of Alaska, yes. You would have to drive through British Columbia, Canada at the very least. For the more isolated part, a plane would be needed, thought you could drive tom a town in Alaska and take a plane from there to the isolated area.

Yes you can. The Alaska hwy has made it easy to drive from Alberta Province to Alaska and it is a very scenic and fun road. Paved all the way. It connects in Alaska to roads that will take you to Fairbanks and Anchorage.
You can also take a ferry from Bellingham WA up the inside passage. It makes stops at about 7 Alaskan ports but only the last 2 have roads that go into mainland Alaska.
I rode my motorcycle up there 2 years ago, and it was fantastic.
Have fun.

You can drive up the coast to Alaska through Canada, but many specific parts of Alaska are not accessible by car. So it would depend on where you wanted to go.
