Was there a real potion in the 1590s that could make someone appear dead for 42 hours, as in Romeo & Juliet?

or was Shakespeare writing science fiction?

    It may be that he knew of uses (or purported uses) of curare from the New World, which has a history as an anesthetic.

    Also, when Shakespeare was writing, there were no EKGs or EEGs. If you "seemed" dead, you were dead.
