Should I report this Ebay buyer?
I posted an item on ebay and somebody won the auction but after they won, they sent me a message asking if they could cancel. Should I open an unpaid item claim or should I cancel as they requested? I'm just not sure how it all quite works.

I would just cancel. Some customers are just unreasonable. If you refuse to cancel the transaction and open an unpaid item case against them, they might just pay but will leave you a neutral or negative feedback. Not all customers are like that but it's not worth the risk. Sending them a cancel transaction request and adding them to your blocked bidder list is probably a better choice.

Don't stir things up in that community,it's best for your credibility to do as asked. Just cancel and relist or offer it to the next highest bid. It will usually tell you that an item is eligable for second chance offer. This has happened to us more than once, these people are just time wasters.

Probably just some dummy who bid on it and either didn't expect to win, or just decided he doesn't want to pay. Best bet (to avoid the hassle) would just be to cancel it for him and re-list the item.

cancel and relist, especially if you hadn't mailed out the item. It will help your credibility as a trustworthy vendor.
