You have to take Shinkansen between Yokohama and Osaka.

There is no way to book its ticket online from another country. But you can buy its ticket in most cases. Don't worry.

You cannot book tickets online outside of Japan in English. You can book tickets inside JR (Japan Railways) stations in Japan.
If you plan on doing a roundtrip between Tokyo/Yokohama and Osaka within 7 days, then YES a JR Pass is good to have. Where else in Japan are you visiting?
From Yokohama you need to go to Shin-Yokohama Station to catch the Shinkansen bullet train to ShinOsaka. At ShinOsaka Station, you need to transfer to a local JR train to OSAKA Station. At Osaka Station you can catch some direct trains to Universal Station, which is the station to USJ.
I recommend you go to tripadvisor.com forums or Japan-guide.com forums for more detailed answers!

You do not need the reservation. Just go to Shin-Yokohama and get on the west-bound Shinkansen. The worst case scenario is you have to sit on the floor between cars if all the seats are filled. However, you will probably get a seat even without a reservation. Yes, a JR pass would work for you.
