What did Jesus mean when he blessed those who believe like Thomas that Jesus is "My Lord and My God"? (John 20:28,29)?
The whole quote, then, since a number of answers haven't bothered.
And Thomas answered and said unto him, My Lord and my God.
Jesus saith unto him, Thomas, because thou hast seen me, thou hast believed: blessed are they that have not seen, and yet have believed.

It means that Jesus really IS Lord and God. Were he not, Jesus would have condemned Thomas as a blasphemer. In fact, he praised Thomas for his recognition of who he really is.

Jesus said that those who didn't see Him, demanding visible proof like Thomas did, would be blessed. Thomas believed because he saw--and probably wouldn't have believed otherwise. Since then, millions of people have believed on and in Jesus for salvation, never having seen Him.
Simon Peter wrote of this, I think, when he said, "Whom having not seen, ye love; in whom, though now ye see him not, yet believing, ye rejoice with joy unspeakable and full of glory (1 Peter 1:8, King James Version)".

Some scholars have viewed this expression as an exclamation of astonishment spoken to Jesus but actually directed to God, his Father. However, others claim the original Greek requires that the words be viewed as being directed to Jesus. Even if this is so, the expression “My Lord and my God” would still have to harmonize with the rest of the inspired Scriptures. Since the record shows that Jesus had previously sent his disciples the message, “I am ascending to my Father and your Father and to my God and your God,” there is no reason for believing that Thomas thought Jesus was the Almighty God. (Joh 20:17) John himself, after recounting Thomas’ encounter with the resurrected Jesus, says of this and similar accounts: “But these have been written down that you may believe that Jesus is the Christ the Son of God, and that, because of believing, you may have life by means of his name.”—Joh 20:30, 31.

Jesus wasn't blessing those that believed like doubting Thomas. He didn't even bless Thomas at that point. He told Thomas and the other disciples that blessed are those that believe without seeing.

It is difficult to determine the intent of an action of someone who did not document his intent at the time of the action but decades after his death some decided it should be documented. Best guess really.
