DNA polymerase has proofreading function but still it makes mistake. However, DNA mismatch repair enzyme corrects the mismatched base pair mistakenly made by DNA polymerase, and reduces error rate down to 1/108 to 1/109. While DNA is replicated, DNA mismatch repair enzyme scans DNA double helix traveling behind DNA polymerase. When it sees mismatch base pair such as A-G, it corrects the mismatched base pair.

The question is how the DNA mismatch repair enzyme knows A is wrong or G is wrong? In other words, how DNA mismatch repair enzyme knows it should be A-T or C-G
