Well first there is the danger of the voyage, there could be bad storms, ship born diseases, contaminated food/water, running out of food, getting lost, could encounter pirates, sea monsters (they believed in sea monsters then) etc

as for once he arrived the natives could be dangerous, there could be diseases, what he is looking for (gold and spices) might not be there after all, also sailors might run off
