I charged $100 for the design and $32.50 for the Vista Print printing. Total is $32.50. Do I only charge tax on the $100?

In California it is the State Board of Equalization. You charge tax on everything you sell. And BTW Vistaprint wasn't $32.50 it was $60 if you expect to make any money. If you don't have a tax ID that makes you a reseller then don't charge tax. Charging tax and keeping it is worse than not charging tax, legally.
You charge for the complete work. IF you are collecting the tax, you do not pay the vendor any taxes. You give him a copy of your permit to collect taxes for his records. You get your license from the Secretary of State [in my state] or the State Internal Revenue service, which permits you to collect taxes.. When you sell your product, you collect the appropriate percentage, and deposit it into your business account. At the end of the quarter you send your Check to the Collector of taxes. Usually, it is the same day as you pay your estimated federal income tax for the next quarter.
