Markham is generally located north of Steeles Avenue (northmost street in Toronto) east of Yonge Street.
Hotels that are located along Highway 7, near the 404 highway should be convenient for you.

If you are looking for a hotel convenient to Markham AND to Toronto itself, the Novotel in North York is a good choice. It's right on the Yonge Street subway and the last time I was there, was a good hotel. On top of which, it's in the heart of the "downtown North York" strip with shops and lots of restaurants.

Residence Inn by Marriott Toronto Markham
Comfort Inn Toronto Northeast

it will depend on budget. Hilton suites can be had for a good deal sometimes too but generally it's higher priced. Anyway check out these links for some decent hotels.
