Ideally, you would do this without a bra on or with a thin one. You'll get the most accurate measurement without extra padding or layers. Grab a good measuring tape. In my opinion, using thicker ones are easier because they dont' twist as much, but it's not a huge deal.

First measure right under your bust. Make sure you hold the measuring tape on your back at the point where your bra is going to be. Don't let the tape go too high or too low. At this measurement, measure tightly. In your case, here you should get a 26 or 27. Add 5 to this number, rounding up to the nearest even number. This is your band size.

Next, measure around your bust measuring tape should be at the same point on your back as it was during the first measurement. Don't let it slide up or down. Here you will want to measure loosely, but not too loose! :) Leave enough room for something about a coke-can size or a fist. In your case, you prabably got a 35 (this will be your bust measurement). So now what you are looking for is the difference between your 2 measurements, so you subtract your band size measurement from your bust measurement. So 35-32=3. This difference of 3 is what you use to calculate your cup size.

a difference of 1 inch is an A

a difference of 2 inches is a B

a difference of 2 inches is a C

a difference of 2 inches is a D and so on...

Keep in mind that this is not really an exact science. It will be more of a guideline. I used to sell bras at GapBody and it usually took the average person tryign on abotu 3 different sizes before they got the best fit. You should really try them on before you buy them as you may need to go up or down a size depending on the style of the bra. And when you try them on, ideally, you'll want the band to fit comfortably when it's on the middle hook to allow some growing or shrinking.

It means that if you took your breasts away, your rib cage measurement would be 32 inches and your cup size is C, which means it is three inches more than 32. So altogether, the measurement around the fullest part of your bust should be 35 inches. HTH : )
