    If you have an unknown sample, you would have to incubate in SIM medium (stands for sulfur, indole, motility). Then the following day add Kovac's reagent and agitate. A cherry-red ring around the top of your test tube is a positive reaction, indicating indole production. The SIM test is only performed to differentiate the family Enterobacteriaciae, which are all Gram - rods. If you don't have a Gram - rod, then this test is of little to no value to you.

    If you just want to know if a specific species produces indole, consult Bergey's manual (the Bible of microbiology!).
