In terms of policy, there is a lot of overlap. In fact many leftists/progressives would say that the Democrats and Republicans are just the left and right wings of the Corporation Party, reflecting the influence of corporate dollars flowing into campaigns and lobbying efforts.
Republicans tend to be more hawkish on war, more likely to support tax cuts, more likely to oppose abortion and gay rights, and more likely to support privatization of social security and other federal programs wherever possible.
Democrats are more likely to support increased taxes on wealthy Americans, support government regulation to improve/protect the environment (such as enhanced emissions and fuel efficiency standards), and public-funded health care, and more likely to vocally oppose torture and warantless wiretaps.
Neither party has shown any real interest in reducing government spending.

Republicans tend to be more conservative, and don't really like taxing or war, or guns. The Democrats tend to be more liberal, are more for taxing, war and guns. This is not a cut and dry answer, but a general overview for the differences. You can also go to Wikipedia and they will be able to explain a lot better than anyone else.
