Sounds somewhat intrusive, but you will need code that runs client side. I assume that you would then want the screenshot sent back to the server or to you?

First and foremost, php is a server-side language. Because of this you will not be able to run the code you need from the client's machine. And last I knew flash would not provide the type of functionality you would need. The only thing php would be good for is using it to handle the screenshot after it has been taken on the client's workstation.

Javascript and VBS are both client side scripting languages that can be embedded within html. I am pretty sure that you would have to work with the clients Win32 API or .NET framework to get the screenshot. If the browser deverlopers have done there job properly and the user does not override the default security settings, the browser should disallow any type of access in this context, making it extremely hard to accomplish this goal. Another possibility would be to create a Java Applet. But the user would have to allow it run first off.
