Can you call a hotel after midnight to make a reservation ?
It's in downtown Toronto and I'm afraid to lose the room to someone else ... the reservation is for next weekend .
I'm sure there would be someone at the front desk at night , but is it appropriate to call at night and book ??

When you are reserving the hotel on line, you can ask them to write down your latest check-in time, to make sure that when you are arriving late, you can still have the room.

If it's a full service hotel, then absolutely it's ok. Marriott, Double Tree, Sheraton, Ramada, Holiday Inn, etc. Go for it.
If it's a mom and pop shop where the person "on duty" is likely napping in the little room that connects to the lobby, then you may be waking that person. Is that wrong? Not really if you are bringing in business... that's part of their job. Should you wait until morning? Well, if the people competing with you for the room will wake the person, then you would lose out.
If you are looking to make a reservation, I recommend you make the call. Have fun!
