I have been working for this company for close to 5 years, in that time I have been offered a promotion 5?
times. Unfortunately, due to family circumstances I had to turn it down. Now I have the capability to accept and there is now an opening. The problem is that I have a new boss and I have a feeling that she is going to either try to talk me out of it or make me look bad so that I don't get it. She is going to have to find someone to feel my current position which is difficult due to the region I am in and it consists of a lot of travel. Has anyone ever been in this position? What did you do?

Yes, I have had selfish bosses that would rather not see me succeed
than to try to replace me. Do all you can to improve your situation.
