The Daddy long legs spider:

They are are actually spiders and like all spiders they do have venom, although the amount isn't enough to do anything to us (unless you have a severe allergy) and they can break the skin pretty much if forced/provoked. This was the one that was on Mythbusters.
This is what we commonly call the Daddy Long Legs (note no spider in this name):

This is not a spider, it only has 6 legs not 8, but this is the one that the myth is based on, not so much the previous spider. These do not have have venom at all, and their chelicerae are used for grasping rather than as fangs in the case of spiders. So that's three counts against the myth, it's not even a spider, no venom, and really can't bite.
On a side note, we only have two truly poisonous spiders in the United States that cause fatalities or severe injury outside of allergic reactions. One of is the black widow and the other is the brown recluse. Those are the only two that the general public would need to be concerned about.

When they say that they are referring to the fact that the daddy long legs has the highest ratio of venom to body mass of any spider, so technically it is the most venomous spider, but it also has a very small body compared to other venoumous spiders. Its teeth are so tiny it can only use it venom on other insects.
The Brazilian wanderer possess more volume of venom then any spider which makes it obviously more deadly.

according to the Guinness Book of Records the world’s most venomous spiders is the Brazilian wandering spider.
This spider is believed to have the most active neurotoxic venom of any living spider. Its venom is so potent that only 0.006mg is sufficient to kill a mouse.

This was tested on the tv show Mythbusters.
The Myth goes that the venom is extremely toxic but their fangs are too short to penetrate human skin.
They gathered specimens and extracted venom and found it wans't true at all.

It's probably the least venomous.

It is not the most poisonous spider but it is very poisonous.
If its fangs were bigger then it would be extremely dangerous.

That was a common rumor, but Mythbusters tested it and they found out it's harmless.
