It takes place in NYC. There are two rival gangs: the Jets (whose leader is Riff) and the Sharks (whose leader is Bernardo). Both attend a dance at a gym, where Riff's best friend Tony and Bernardo's sister Maria meet and fall in love. After the dance (and after Bernardo tells Tony to stay away from Maria) the two gangs decide on an all-out rumble (or fight). Meanwhile, Tony goes to meet Maria at her balcony (it looks like a fire escape). They sing a song about their love, kiss, and Maria tells Tony to meet her at the bridal shop where she works the next day. Oh, and before the gangs decide on the rumble, the Sharks sing "America" which compared Puerto Rico to America, and the Jets sing "Cool" in Doc's Drugstore to release their steam. The next day at the bridal shop, Tony comes in while Anita (Bernardo's girlfriend) is still there. She is mad at Maria, but doesn't tell. Tony and Maria "get married" while singing "One Hand, One Heart." In the next scene, the Jets, Sharks, Anita, Maria, and Tony are all singing about what will happen tonight in the "Tonight Quintet." The final scene of Act 1 is the rumble, where a simple fist fight between Diseal (one of the Jets) and Bernardo ends up becoming a knife fight between Riff and Bernardo. Tony comes and tries to stop it due to Maria's wishes, but fails. Bernardo accidently kills Riff after stabbing him, and then Tony gets really mad and kills Bernardo. All the Jets and Sharks start fighting but stop when they hear the police coming.

Act 2 starts with Maria singing "I Feel Pretty" with her friends, and when they leave, Chino (the guy Maria is supposed to marry) comes, and tells her what happened, but she doesn't believe him. Then Tony comes, asking Maria for forgivness, which she does. They plan to run away. The Jets, trying to cheer up after their leader's death, make fun of political figures. Next, Anita sings to Maria about how she should not be dating Tony, but Maria sing back to her, and by the end of the song, Anita is on their side. She reveals that Chino has a gun and is going to kill Tony. In the next scene, Anita goes to Doc's Drugstore (where Tony is) to tell him to look out, when she runs into the Jets, who taunt her and almost rape her. She gets really mad, and ends up telling them that Chino found out about Tony and Maria's love and killed Maria. The Jets then tell Tony, who decides to let Chino kill him because he took away his love. Tony runs the streets, and runs into Maria. They hug, but Chino finds them and shoots Tony, who dies. All the Jets and Sharks come on, and 2 Jets and 2 Sharks carry Tony's body off stage, symbolizing the end of war between the two gangs.
