how does low pH interfere with an enzyme active site? a high pH?

    There is generally only one pH at which the active site is the optimum shape to hold the substrate. Any change in the pH (increase or decrease) will cause a distortion of this site, making it less effective. The greater the change, the more the site is distorted, hence the typical bell shaped curve when plotting rate of reaction against pH.
