Mystical space typhoon (MST) can't negate the effect of a card, unless the card is continuous. It can't negate a single turn card such as sakuretsu armor, magical cylinders, etc. Its best use if your opponent plays a "facedown" cause it would be destroyed. If your opponent activates a card (unless continous) it will NOT be negated.
Book of moon can be chained to almost anything. It changes a monster on the field ionto face down position. So if your going to be attacked, you can activate it (as long as its face down) and save yourself from the attack. If there are cards equiped to a monster, you can use book of moon to place the monster face down, and the cards (since they can't be equiped anymore) are destroyed. There's no tricks, it just works like that. Mst destroys cards, not negates them, there for only useful on facedowns or continuous. Facedowns CAN still be activated even if you use mst unless it was the first turn they were played. And book of moon forces the monster of your choice into facedown position, making any cards associated with this monster, unaffected and destroyed. Generally speaking.
Hope I helped :) happy dueling!

1. The monster would be placed face-down, but any priority effects would still resolve normally.
2. MST does not "negate" the spell/trap card, therefore the effect will still resolve normally (unless it is a continuous spell/trap card that must remain face-up).
Use MST during your opponent's end phase so they cannot chain your target.
