Thin layer chromatography help please!?

in general, we assume that in a mixture, each component will show the same rf value it shows when it is run alone. sometimes the presence of a large amount of a second component will affect the rf value of a component of interest. suggest why this behavior is found?

2.) how could you use TLC as an aid in finding out how to seperate a large quantity of material (5 g) by column chromatography?

    1) If both components exhibit similar polarity, the second, if in great excess, will influence the primary analyte to travel a distance similar to itself, thus changing the Rf.

    2) Using eluting solvents of non-polar gradually increasing to more polar to get good separation is the key here. Once this is underway, take multiple fractions of equal volume and use TLC to see where the "breaks" are between different compounds. So you would run TLC on each fraction and combine all fractions that have the same spot on the plate.
