    Loading dye is added to every sample you plan on running. If the standard is commercially obtained, it may already have loading dye in it. The dye serves two functions: (1) to monitor the progress of electrophoresis, and (2) the loading dye is dense, and allows the sample to sink into the well. Without it, you can't load the sample.

    Yeah, it has to be. Otherwise, you wouldn't be able to see it in the gel electrophoresis.
