Should I put export my video from iMovie in 540p (6 gig), 720p (10 gig) or 1080p (16 gig).?
It s 2 hours long (seminar). I will be putting the video on YouTube and I m worried that it will load slow if I use 1080p. I ve also heard Samsung devices struggle with 720p? Is there a huge amount of difference between the 3 quality wise? Thank you.

You should upload the best version you have to YouTube. YouTube will process the video and create several versions for streaming. They stream the most suitable version for the speed of the connection being used so a faster connection will get an HD 720 or 1080p version and a slow connection will get a 480 or worse version. You should use the MP4/H.264 settings for best result to file size.
You will need to check that your YouTube account allows for a 2 hour video - you may need to edit it down in to several shorter parts.

Yes, there is a visual difference among the three. 720p is (still, unfortunately) an acceptable visual rendering for video.
