I have an HVAC question. I need to separate a part of my house from the heating system, as I am doing renovations. How could this be done?

It is February. I found some mold growing under my old wallpaper that is caused by a roof leak. I need to repair immediately and continue living in the house, if possible. My idea is to seal off the living room. What is the most effective way to seal off an open doorway? How does the HVAC system read the temperature of the house? I need to make sure it isn't measuring the temperature of the living room as we will be ripping walls and insulation out. Does this even sound feasible?

Update:

I have a 1960's ranch style house. I know the industrial system at my work uses temperature sensors inside the duct-work. I'm not sure how residential heating systems work. The thermostat is not located in the living room. Is it safe to assume that my furnace will not read the temperature of the ducts leading to my living room?

    It's safe to assume. The thermostat has the internal sensor and reads the temperature at the thermostat.

    Close the vent in the living room. If it doesn't have louvers then cover with plastic. Make a door out of 3mil plastic so seal off the room.
