What comes immediately to mind is a stroke.
This works on two counts:
1. A stroke affecting motor functions of the larynx preventing vocal chord use or the mouth/tongue preventing the formation of sounds used in word formation.
2. A stroke affecting the speech center of the brain.
While it is not a disease, an infant who is born 100% deaf will be mute or, at best, VERY unintelligible. This is because talking is learned by imitation; a child who hears absolutely nothing, will not have any idea about what using the vocal chords does or how to use them.

It's not a "disease" but losing your teeth due to not brushing or flossing & rotting means no teeth and that can make it hard to talk without fake teeth or denchers (I don't know how dencher is spelt).

smoking a crazy excessive amount can destroy your voice completely and leave you mute (besides groaning sounds) and in need of some machine thing you place on your throat and it makes an artificial voice based on the vibrations.
