As hilarious as that would be I think they'd have a hissy fit because the character would have to have some giveaway that they are transsexual or nobody would know. Not many people read the backstory of existing characters but they have things about them so you know what they are meant to be.

That would be cool, but only if it's what the people making the game want to do. And it would be ideal if the fact they're trans isn't like, used as a selling point or anything. Just make them like any other character, really.

The question is, how would the character's gender identity be relevant to the story of the game? If it's some pertinent detail to the character's back story, then fine. If it's a secondary/background character who appears to be trans but there is no emphasis on it, then fine. But if the main character or a secondary character is specifically pointed out as being trans when it has nothing to do with the game or the story, what's the point? The same goes for a character's sexual orientation. For example, there was one game in recent years (the name escapes me...it was set on a space station where everything went wrong and you're investigating the crew's disappearance), which had a same-sex couple on board and another gay character whose same-sex spouse was back on Earth, out of a total of maybe 8 crew members. That one was somewhat borderline because while it was treated as a somewhat unemphasized detail to the story, like there are these characters and some just happen to be homosexual, the high ratio of homosexual characters compared to the average population seemed to be pandering to the GLBT community with an unrealistic amount of diversity. I say somewhat unemphasized because while it was treated as "they just happen to be gay", that detail kept coming up time after time as the story progressed.
So when scripting a video game and casting the characters, the game designer has to be careful to have some details and back story of a character without details irrelevant to the story or game play becoming unnecessarily overemphasized, whether the detail has to do with gender identity, sexual orientation, race, nationality, or whatever else. To answer your question, there's no reason there shouldn't be transgender video game characters, but there should also be a reason for that detail to be included as well.

Anything to add a bit more colour and adventure to the video game is fine by me. I'm still waiting for a superhero who is a closet crossdresser.

There are as many as you like, because any player character whose gender differs from the birth-assigned gender of the player is trans, and in fact that phenomenon is probably instrumental in more people coming out as gender incongruent in the past couple of decades. I used to play as a female character years before I decided to transition.
