In what ways is a lion “big-hearted”?
Is this merely according to symbolism and mythology or is it something that is scientifically proven in a lion’s temperament?

It's simply a symbol of strength.
Lions are no more big-hearted - literally or figuratively - than any other animal.

It's symbolic, likely because their chest is so puffed out with all that hair.
In reality, lions are quite fearsome, even to their own kind. An alpha adult will kill cubs if he finds them, because the cub may grow up to compete with him for leadership of the pride. Lionesses have to protect their cubs from males until they have grown enough to care for themselves (about nine months).
