It all depends who your ISP is. If you are talking about Yahoo, YES it does matter; it improves the Spam filters and the spam will no longer come in your mail box but in the Yahoo balk folder. Just leave it there and it will by deleted automatically.
Go to Yahoo mail options and set it up any way you want.

You can report till the cows come home. Spammers have many tricks to change where a spam is coming from. One of those tricks is to infect millions of unsuspecting computers with viruses that work as 'spambots', sending out spam emails and hiding the actual sender of the spam. The ONLY way that I have found to completely eliminate spam is a free email service that I found. http://www.enterto.com/ uses channels. If someone sells your email address to a spammer you just delete that channel and you cannot get any more spam.

I have no idea. I always assumed the system aggregates what users say about email addresses and builds new spam lists based on that.
But since it takes exactly the same amount of effort to mark an email as spam as it does to delete it, does it even matter?

when you report the e-mail as spam... the mail server company will monitor the e-mail address that you said is SPAM and if it really is SPAM e-mail, it will block it from sending.

if you want to spend all your time doing that go ahead. the rest of us will be having fun while you are working your butt off reporting them.
