I have a 1999 Ford E-450 Super Duty van?

as the basis for my motor home. It has a 414 V-10 and automatic tranny. There is a button in the end of the shifter knob. What the heck is it? Does it engage the OD, or what?

Well like the first guy said it turns on / off the od its a ground switch and it also flashes when there is a transmission code. When it starts flashing it could be electrical malfunction or gear selector switch out of range so it has many functions.hope its not flashing.

I'm not positive, but it probably keeps the overdrive from kicking in when you're towing a trailor. On an automatic tranny the button is on the end of the shifter. Try pushing the button and see if a message appears on the dash near the turn signal indicator.

It's the OD button to turn the OD on and off.

To turn over drive off or on. It will light up when you have over drive OFF.
