Is your mouse USB or PS/2 ? If it's one - try the other.
If USB - unplug all other USB devices, then plug the mouse in after the computer has booted.
if PS/2 - unplug the mouse and be sure that there aren't any pins bent down. It's also not uncommon for the little plastic tooth in the center to break off inside connectors.
Ctrl ESC = Hitting start button
Windows Key + Pause/Break Key = My Computer Properties
Hope these shortcuts help a bit while you are troubleshooting anyway.
Make sure no USB controllers are disabled or malfunctioning - or that the PS/2 controller is present and not flagged.
Both of these connectors are breakable - try the usb in a different port - or a different group of ports. PS/2 - you can only plug into the one hole.
Try a completely different - and known good mouse - and try your existing mice on the other computer.
Best of luck.

If it's a PS2 mouse port and you plugged a mouse in without unplugging your power supply or turning if off at the rocker switch on back you may of fried your mouse port. With your puter shut down there is still power to the PS2 ports. Spend 10 bucks and buy a USB mouse. One of the most common mistakes people do is plugging mice and keyboards into PS2 ports frying the ports. They can do it ok 10 times but then, if they're not lined up right POOF goes the port.

Might be a broken port. If you are using a PS/2 mouse, try a USB one, likewise if the mouse is USB.

if you plug in the mouse after you turn on the computer, it won't work. did you try turning the computer on and off again?
the usb port or hook up your using could be broken, try using different plug-in spots.
hope it helps!!
