Are there any words that can ONLY be defined by using itself?

No. Language is a series of relationships and contextual understanding. The closest you can get to your challenge is, if you look into the language of music you could say an eighth note is the same as two sixteenths tied together... now, I know that I've defined an eighth note as something other then itself, however... they sound exactly the same! In a language like english, if you described a word using other words they would not sound the same.
On the flip side. YES, always. To describe anything by something besides itself is to describe itself.

In fact colors can be defined by the number of light waves propagating throughout space.
I.e. red is light exhibiting a wavelength range of roughly 625–750 nm.
To answer your question - I don`t think so. You placed emphasis on "Only," - if that`s the case I don't think there is a word that can "Only" be defined using itself. Language is such a cohesive blend of thoughts and emotions we can always define words with other words or a by a set of words.

Any definition should be able to be reduced down to a two word definition such that neither of the two defining words is approximately the same as the word defined or the same, and that the definition is sufficiently accurate.
One only needs to check some words: 1) abstract concepts generally considered difficult to define (opposite, negate, thought, etc...), 2) basic words (walk, go, be, do...). Or we could take the some 8,000 basic words and reduce them to an inventory of 3,000 which need to be defined to test the theory, and then define them.
I am willing to work on such a project to answer this question. This project needs 4 phases:
1) define parameters and nature of work of the project .
2) decide upon the inventory of words to define.
3) work on defining the words until all the heads of the project can agree on the adequacy of the definitions suggested.
4) publish the work.
Create a Yahoo group for it, invite some serious minds, and email me when you do.

Words are by itself a definition. When an object or concept is named, that word becomes the definition of the object.
It must be noted that what exist is the concept, the word was created in the human mind or invented for that concept and not the other way around.
Hence, the color black is called "black" in some language, "hitam" in other languages, "heisu" in other languages and "itim" in my language. The "blackness" of black or its being retain itself despite the different words we use for it.
