Restriction Enzymes.......?
How can you estimate when restriction enzymes will cut, and how can you predict the frequency??? Please help, exam tomorrow!!!

you cant exactly estimate when or the frequency unless you have the sequence of the organism
for example,
the enzyme HindIII recognizes the sequence AAGCTT
whenever it sees this in the sequence of the organism, it will cut it there
so you need the DNA sequence to check
i do this all the time!

Look at the what nucleotides the restriction enzyme has because it will bind with hydrogen bonds to the sequence where it cuts. So if you know the nucleotides of the enzyme you will be able to determine where it cuts.
To determine the frequency just calculate the percentage that that specific sequence would occur. For example if it was a 4 n.t. sequence: ATGGTA then the frequency would be (1/4)^6. Because there are only four different nucleotides.
