George Carlin is the best stand-up comedienne I know. His jokes not only make you laugh, but they'll make you think as well. TV-wise, I think Tina Fey is a pretty decent choice for a female comedienne, then there's Simon Pegg (funny Brit guy from Hot Fuzz)

Jon Dore's standup was very funny. I agree though that Jim Gaffigan is the best, loved watching him on Conan O'brien all the time. Jon Lajoie makes really funny videos on youtube. Most are comedic songs or "ads".
