Depends on the system in place. In the commonwealth, the President is often the head of state and the Prime Minister is the elected leader of a nation, most often in a parliamentary system where each constituency elects a member of parliament who in turn elects the prime minister.
A president can be directly elected by the people in a democracy, but some nations have other ways of electing a president.

A prime minister is the "head of government", meaning that they control the country's administration and executive. The president is the "head of state", which is the person who is formally in charge of the country. Most presidents are also the head of government as well (in countries that have a prime minister, the head of state is usually someone else and is more ceremonial). The exact power between the two varies depending on country, but generally presidents have more power. Prime ministers rule because their political party has a majority in that country's legislature, and hence they need the support of the legislature to keep going. Presidents do not. So for instance, the Canadian PM gets his power because his party has the most seats in the Canadian House of Commons. If he loses the support of the house he has to resign. The US president, however, doesn't have to worry about that (you'll note that Obama is still the president even though the Republicans control the house of representatives) and can keep going until the next election. Some countries have both a president and a PM. If they do, usually the PM is the one who effectively runs things and the president is a ceremonial figure. If they only have one, whichever one they have is usually the one who runs the show.

First you have to develop the ruling regime, in a democracy a president is regarded as a high authority position and has a lot of rights, the prime minister in this case is more like the spokesman of the ministers from one side, and a right-hand in diplomatic affairs. Global politics are usually set by the president and followed strictly by the prime minister however most democratic presidents actually follow the ministers decisions most of the time giving the prime minister an indirect power to impose some politics.
In a republic the power is usually split amongst the president and the prime minister in a more horizontal way, the prime minister in this case is a major part of the global political scheme and usually has much more profound roles to play especially in international affairs.
Republic laws change radically from one country to the other but in most of them the parliament has a great authority granting a big push to the role of prime minister.
In modern republic-monarchies (ie the United Kingdom), the prime minister holds the absolute power like a president and the prime minister combined while the King / Queen are more of a cultural symbols to the state. The prime minister in this case still answers closely to the ministers who have the right to relieve him of his duties.
This does not apply to strict monarchies like in Saudi Arabia where the king is the sole ruler.

A prime minister rules with a government eg. he doesnt rule by himself/herself and makes decision with help of his party but a president makes all decisions on his own and controls his country by himself

A President is the head of state. He/she can be powerful (USA, France) or not (Israel, India).
The Prime Minister leads the gov't with a President or Monach above as the head of state with great or little or no powers.
