Linux is all about choice, which is it's strength, and its biggest weakness. There isn't a one size fits all answer. Without knowing which distro (Fedora, Gentoo, Ubuntu) or anything about your hardware, it can't be answered.

Try checking the documentation or online wiki pages for the distro you are using. If you can't find your answer, go to the forum that is specific to your distro, but be warned, it is common to run into butt heads in there that will call you a n00b and tell you to read the docs. So don't post your question as you did here, but ask something like "I can't find a document that tells me how to use the CLI to configure my wireless NIC to connect to an access point" and tell them where you have already looked. Chances are somebody will be able to find a Howto for you.
