Yes and no. In the first one you were Demond Miles, and you were hooked up to a machine that made you relive your ancestors, Altair's, memories.
In Assassin's Creed 2, you still play as Desmond miles, but the machine makes you a completely unrelated ancestor.
So if your just play the game to be the assassin, then it doesn't matter, but the plot concerning Desmond Miles will make no sense.

I am pretty sure that the Assassins Creed 2 game will not have anything to do with the first one, other then the fact that its about assassins :). Assassins creed 2 will have a new assassin Named Ezio. Also the timeline are in two different times.
