He lies about helping Roderigo win over Desdemona for Roderigo's gain, and not his own. He also lies about his suspicions about Desdemona being unfaithful to Othello with Cassio, Othello's lieutenant. And nobody realises how evil he is, and because Othello trusts him so much and thinks of him as "honest", Othello completely believes Iago's "suspicions" to be true because he thinks that Iago is knowledgeable and wordly.

Iago lies about Desdemona, Othello's wife, sleeping with Cassio, Othello's subordinate. Othello believes him.

That is the point of the entire play. Othello believes the lie and this leads to some.... unfortunate consequences.
