Would it be:

The largest problem with Canada’s upper chamber is the Senators that it is actually comprised of.

OR

The largest problem with Canada’s upper chamber are the Senators that it is actually comprised of.

    The largest problem IS .... Actually, I'd turn it around and get rid of the dangling 'of'.

    The Senators that actually comprise it are the largest problem with Canada's upper chamber.

    The subject is "problem," which is singular. In this case then, use "is" with the singular
