Can I cross the border to mail something?
I have a big box to mail to someone in Canada, but I live 100 miles into the USA. It's expensive to mail it from my town. Could I drive across the border and mail it from there?
I'm up near the border every month for work, so crossing isn't terribly inconvenient. Also, I'd be crossing at Niagara Falls, and I could turn it into a nice day-trip.

If you mail your package in Canada, you have to pay Canada Post or FedEx or whoever up here to deliver it. Have you compared what the cost from USPS is compared to Canada Post for delivery of that parcel?
You mentioned that you are up near the border every month for work, but I doubt you are right at the border, so you have to justify the cost of driving at least a few miles to the border, waiting in line to cross (usually anywhere from 15 to 60 minutes), finding the nearest Canadian post office to get your package submitted to the parcel post delivery to be delivered, and then waiting in line again to cross back into USA. You also have to have your passport, NEXUS car, or Enhanced driver's license on you or they might not let you cross.
Canada's customs officers will also want to know the contents of the parcel and the value of it, because there may be duty and taxes to be paid on it, which you would have to pay for anything over CAD$20.
And crossing at Niagara Falls, I believe the bridge has a toll on it.
I've never heard anyone who did this, but it is certainly possible to be done. When crossing, if you are going to spend time as a tourist at Niagara Falls, then you can indicate that as the primary purpose of your visit. Dropping off a parcel at the post office is secondary. But legally, you must advise them of the package, otherwise you could face some sort of fines or even smuggling charges.

You could do that if you wanted. You will still have to take the box through customs though. Personally, by the time I'd factor in the drive to Canada, the time it takes, the cost of gas, the hassle of crossing the boarder......it would spend more then if I just mailed it from home and was done with it.

Sure.
You obviously need a passport, take the time to drive and wait in line through the border crossings, explain to US officials what you are doing (and hence pay duties depending upon the value of the item), then find a US post office near the border, and go through the trouble of mailing it in the US.
If you pay yourself even minimum wage for your time... it can't be cost effective. However, nothing prevents you from doing it.

Do you have a passport? Without one, you won't be crossing the border.
You have to declare goods brought into Canada and might have to pay duty on them.

Horton of course you can, as long as you have the right travel documents. Your crossing an International boarder my friend, not going to another state.

so the cost of gas and wear and tear on the car for 200 miles is going to be less? have at it. remember to claim it at the crossing.
