No, they are not the same.
¿Cómo estas? is correct.
¿Cómo son tú? is wrong.
Son is third person plural, where tú is second person singular, so the verb doesn't match the subject.
Although it is possible to say ¿cómo eres tú? [what are you like?], it doesn't mean the same thing as ¿cómo estas? [how are you?]
