To do something heartily is to do it with enthusiasm as if you really care. Drink up means drink all you have. Hearties seems to be a naval term of endearment from a higher rand of seaman such as a mate ot a busun to his crew. So 'drink up my hearties' would probably be said after the bosun had bought the drinks and now it was time to go back to the ship.

i don't think it's "hearties" either.
