Could anyone explain what the abovemention is all about. And also if possible in what way can column chromatography be fitted into this???

It is a method for studying complexation reactions. E.g. you can have samples of DNA with varying concentrations of a DNA binding protein and you can see if the protein binds to that sequence and have a (semi-)quantitative idea of the strength of the binding.
Column chromatography is usually used to purify the protein that you are going to use in the assay. Otherwise you could use the gel shift assay to find for example which DNA sequence (out of a pool of certain sequences) is best for binding the protein and then conjugate it to the resin of a column and use it for affinity chomatography purification of the protein instead of purifying through tag-antibody affinity chromatography.
