what brand of radio? model?
You are sure that the CTCSS is correct.
Do you hear anyone?
Are you sure the repeater is on the air?
Have you verified that the radio is transmitting?? Did you use a scanner, a freq counter?
I would check the TX using a VHF watt meter.
Check your receiver with another radio... have you got a hand held or a mobile to listen with?

You are a ham, you are supposed to know how radios work.

Invite a friend over and try to transmit simplex to them, start from there. Also, are you sure you didn't activate the sqelch tone instead of the transmit tone? Are you transmitting and receiving on the right frequency?
