What is the best way to quit a job?
I started in an occupation that had nothing to do with what I took in Univeristy just because there were no other jobs out there, and I'm finding that I absolutely can't stand it. It's not at all what my heart wants to do and I go in miserable every day. It's to the point where I can't just put up with it until something better comes along.
The people I work with really like me and think I'm doing well and like the job. Unfortunately, I hate it! What is the best way to quit? When is the best time to quit? I desperately need to walk away with some references.

Use your dislike towards your current job as motivation to find a new one. Set a standard that you're looking to meet in a new job, you may want the next job you work to be something to last you in the long term. A resume full of short periods with different employers shows someone who's hiring that you often like to jump ship. Once you find a job in your field that you wish to accept and offer for, give your current employer notice and keep them in good terms. Since they like you, it sounds like they could make a good reference in the future, though they may not wish to lose you now, so using them as an immediate reference may not work too well.

First, get another job lined up before doing anything else, the job market is sketchy at best, and its better to have a job you don't particularly like than being without one. Most employers will allow you to state when you will be available to start working, and many will not higher somebody who doesn't currently have a job.
Second, once you get a job lined up you need to write up an intent of two week notice, this will notify your boss and employer that you are leaving the company, generally these are a letter with strait forward statements about your intentions of leaving and how long you will be continuing to work in your current position, usually 2 weeks, most employers require this to be filed if you ever want to use them as a job reference, up and quitting can be considered as a serious grievance against the company and you will be considered not hireable because you left the company on bad terms, the 2 weeks gives them enough time to line up somebody to replace you.

Type up a formal letter of resignation providing the employer 2 weeks notice. Hand it to your supervisor on Monday. Offer to help train/mentor your successor if they want you to do that. How much you tell your supervisor about your reasons is up to you.
If you absolutely need a job, have the letter typed, ready to go, but undated. Do not hand it in Monday. Spend the next few weeks polishing your resume and create a template for your cover letter. Once you are comfortable, start job hunting. If someone offers you a job, tell them you can start in 2 weeks, get the resignation letter, type in the date and hand it in.

put your 2 weeks in just to let you know it looks really bad if you quit and try to find a job

First get a new job, then hand in your notice and say thanks, if they don't like it...tough!
