First, whom has legal custody?
If your wife has legal custody, then I assume that a criminal complaint was sworn out. Therefore if this is an issue where of unauthorized custody by a non-custodial parent then I would call local law enforcement.
Now if you want to get a paternity test you should go ahead and go through the courts. If your wife is willing to testify, this should give weight to the claim.
You have several states involved: Indiana, Kentucky, and West Virginia. In which state did the Juvenile and Domestic Relations Court award custody? Start with that state! Once a court makes a decision, you can contact the law enforcement and/or courts in another state and get them to move on it.
I hope that this helps.
Good luck.

get local lawyer to go to court and request the DNA test. If it is your kid, you then file for full custody.
Good luck.
