Before all of the Black Ops fan boys come on here spamming you to buy Black Ops, I will list the reason why each of them might be a good choice.
Black Ops will have much more players online, many more than AC:Brotherhood will probably ever have. It also has better graphics than AC:B, and much more variation (more weapons, areas, etc.). Although remember that Black Ops has a linear campaign mode, and a limited number of multiplayer maps (but i'm sure many, many more will be added through DLC) - But remember that it is a first person shooter, which personally all feel too similar to each other and never have much change from the previous installments/other fps games.
Assassin's Creed on the other hand, will again have a limited number of multiplayer maps (also DLC possible), and nowhere near as many weapons as COD:BO - but the campaign mode is open-world, unlike putting you on a predetermined route such as COD single player. It also has multiplayer added, a new feature for the AC games, and as it's release was only recently many people will still be playing it. Assassin's Creed is also much more story-driven (imo) than COD.
Personally I would suggest COD:BO is you enjoy multiplayer more than campaign modes, and AC:B if you prefer the opposite. Although keeping up with Activision's 'Call of Duty game every year' obsession, COD:Modern Warfare 3 is slated to be released in 2011, AC might have a longer multiplayer following than Black Ops will. COD:MW3 will again be a (slight) upgrade of Black Ops, and will may well be worth waiting for instead of buying the current title of the series.

Assassin's Creed Brotherhood, easily.
Black Ops is a huge mess at the moment, the online is an absolute shambles and the treyarch dev team are no where near fixing it. Definitely get black ops at some point because it is a good game, just leave it a few months so that all the kinks are completely ironed out.
Assassins Creed Brotherhood is a brilliant game. The single player is brilliant and improves on AC2 a lot. The multiplayer is a really refreshing and fun experience as well and it will keep you entertained for hours.

I would go with Assassin's Creed Brotherhood because it has way better graphics than Black Ops. I am a big fan of Call of Duty games but this is by far the worse. The graphics are bad, guns look like plastic and fake, killstreaks that you use in mulitplayer don't go toward other killstreaks. The only thing to look forward to is Zombies but i just dont think its worth it. I have not played AC Brotherhood but i do own and have beaten AC one and i am thinking about buying brotherhood.

Black Ops obviously. The game has amazing graphics and is the best COD that has ever come out. And instead of unlocking attachments, colors, tiles, and guns, you buy them!!! It's way easier that way.

I'd go with Assassin's Creed, not a fan of Call of Duty.
HOWEVER
If you're willing to take a third option, go with Dwarf Fortress.

black ops in my opinion was a let down for all the hardcore COD players out there AC3 is a must buy
