Nice to have that kind of set up.

If the duct is delivering air into the room, they call the adjustable vent a register. If its taking the air displaced in the room and pulling it back down to the furnace, its called a return. So since you can open and close them, lets assume they are the registers. Just remember that warm air rises and cooler air falls and you can figure out how to set them again later if you forget. Since warm air rises, close the upper register in the winter so that the area close to your feet gets heated first, and then the warm air rises up. Reverse the logic in the summer letting the cold air in through the higher register, and it will fall in the room and go back through the return ducts near the floor.

Do you have vents in the room with no controls to open and shut them? If so, those are the cold air returns. If not, check when the furnace or AC are blowing air if all of the vents blow air when opened, or if only half of them do. If only half do, you'll want to reverse the settings of the ones that don't blow air from the ones that do.

This is because the air pushed into the room will displace air that is not at the desired temperature any longer. That air goes back through the return ducts to the furnace or AC system to be heated or cooled and sent back up.

Just leave the both open. If this is the only source of returns I would advice to leave the both open to maximize as much return that possible.

usually there is in basic terms one return air grill interior the abode. this is usually a huge grill approximately 18" or 20" x 24" or 30" or so. My 2,500 squarefeet abode has one return, as did my previous abode, and my early life abode. the finest thank you to tell it as quickly as the gadget is on, carry a splash tissue close to the grill. you will understand good away no count if or no longer this is a furnish grill or return grill.
