Coffee ground will not give you any noticeable advantage. Careful with the heating pad as it will dry out the soil fast. Keep the planting medium moist until sprouting.

coffee grinds are definitely good for some soils to help with growth BUT not for every one. You should test your soil and see if it lacks certain elements - if it is low in nitrogen? Also make sure to put the seeds in direct sun for 6 hours a day or a bit more. Once the first 2 real leaves pop up (not the first two you see but the next 2 - so when they have 4 leaves all together), take the plastic off the cup. Let them grow a bit more, watering only when the seed starter is dry, until they have a few more leaves and seem taller. Then transplant them to a larger pot or put them in the ground (only if it is over 50 degrees at night). They should do just fine if you follow these rules. I have been growing tomatoes from seed since 1995.

Tomatoes will germinate without a heating pad, so be very careful not to over heat the soil. The best tip I can give you is to put the new plants in a bigger pot (say, five gallons or more) just as soon as they develop their first true leaves. They will grow just a bit, then just sit there doing nothing until they die, if you leave them in a four inch pot. No matter what you feed them. Coffee grounds work best in a compost pile or lightly worked into the soil of a garden. They will do little or nothing for your potted plants.

You got it right to start. Just be sure to grow the plants under proper grow lights or direct sun or the plants will become spindly. I presume you will transfer the three plants into the garden when it's time. After the plants emerge with one or more true leaves, remove the plastic cover. It would be a good idea to soil test your garden to see if you need to add lime. That will prevent blossom end rot. In most cases it will not hurt to add garden lime to your soil without the test, but that remains on how professional you want to be. Coffee grounds will not contribute much to your yield, but as an amendment to your soil, it can only benefit your soil structure. Do not add too much. Good luck with your experiment.
